Gryphon™ pCHAC Retrovirus
Retroviruses are considered the easiest and fastest means to deliver genes stably to mammalian cells. Allele has upgraded its retrovirus packaging from the previously offered Phoenix packaging system. The system provides either an Ecotropic packaging system (for dividing murine or rat cells) or an Amphotropic system (for dividing cells of most mammalian species, including human). We have upgraded our custom packaging method to a more efficient system that routinely results in a titer of 10^8 or higher. We have stopped offering the previously known Phoenix amphotropic and ecotropic cells. A similar Ecotropic and Amphotopic packaging cell line, Phoenix2, originated from different sources that are currently available.

Sapphire™ Baculovirus
The baculovirus expression system (BVES) provides a high-yield and low-cost platform for expressing functional eukaryotic proteins. Most eukaryote-specific post-translational modifications, such as signal cleavage, phosphorylation, glycosylation, and disulfide bond formation can occur in baculovirus-infected insect cells. Allele Sapphire™ Baculovirus DNA was specifically designed for efficient recombination with a polyhedrin promoter-based transfer vector by a simple co-transfection step.
- Virtually 100% recombination efficiency, which eliminates the time-consuming plaque purification step.
- A protein disulfide isomerase gene was inserted at the p10 locus of the virus to ensure proper protein disulfide bond formation.
- The Sapphire™ Baculovirus DNA is compatible with most of the commercial polyhedrin-based transfer vectors.

Lentivirus/Retrovirus for Gene Expression
Allele´s Custom Service Division brought to the market advanced lentivirus and retrovirus packaging services. They are highlighted by a consistent high titer, large delivery viral counts, and a 100% success rate. Typically, lentiviruses are packaged by co-transfecting a number of helper plasmids and in most cases amounting to 10^6TU (transduction units)/ml which is considered a good titer yield. The proprietary packaging technologies that are used to perform Allele's virus packaging services are unique and highly efficient, yielding 10^8 to 10^9TU/ml; without any concentrating steps. With a concentration step, however, 10^9 to 10^10TU/ml can be achieved which is particularly useful for in vivo uses.

Description: Amplification/isolation of the gene of interest out of a customer-supplied vector and subcloning it into recombinant Baculovirus transfer vector. Verification of the subcloned gene by restriction digest and sequencing. Preparation of the recombinant vector DNA suitable for co-transfection with baculovirus DNA.
Phase II: Cat# ABP-CS-PEBV002
Description: Cotransfection of recombinant Baculovirus transfer vector containing gene of interest with 3 different linearized Baculovirus DNAs. Recombinant clones will be identified and amplified. The customer will choose the best recombinant baculovirus for the protein expression.

Description: Amplification of the chosen recombinant Baculovirus and titration of final high-titer stock by end point dilution

Protein purification using appropriate affinity resin for 6xHis or GST-tagged recombinant proteins. Purification can be performed either under native or denatured conditions (as desired by the customer). SDS Page and Western Blot will be performed for quality testing.
