Prostaglandin D2 (PGD2) is biosynthesized in the brain by a soluble, 26 kDa glutathione-independent lipocalin-type PGD2 synthase. This PGD2 accumulates in the cerebrospinal fluid (CSF), where it induces physiologic sleep in rats and humans. PGD2 is also synthesized in mast cells and leukocytes by a cellular, myeloid-type, glutathione-dependent PGD synthase. This PGD2 which is formed in the intracellular and vascular compartments is rapidly metabolized to 11beta-PGF2alpha and other metabolites. Measurement of the parent eicosanoid PGD2 is appropriate in cell culture lysates and in CSF, where concentrations of several hundred pg/ml have been measured. Cayman's PGD2 EIA Kit is a competitive assay that can be used for quantification of PGD2 in cell culture lysates and enzymatic reactions.

This EIA offers the convenience of a fast assay with a two hour incubation time and one hour development time, while still achieving an B/B0 value (50% IC50) of approximately 1,300 pg/ml and a detection limit (80% IC50) of approximately 350 pg/ml.

Description:
Many lipids have a free carboxylic acid group that typically requires a weak acid mobile phase to ensure retention by HPLC. Unfortunately, the presence of the acid in the mobile phase also suppresses anion formation and often leads to poor electrospray ionization and poor MS sensitivity. The Cayman’s AMP+TM Mass Spectrometry Kit contains a positively charged reagent (AMP+) that can be coupled quantitatively to free carboxylic acids through an amide bond linkage. The resulting derivatized product thus contains a positively charged group that shows improved electrospray ionization and consequently improved MS sensitivity. Cayman’s derivatization reagent is stable to MS/MS fragmentation and thus isobaric derivatives undergo compound-specific fragmentation to give unique fragment ions.

The active forms of caspases are generated by several stimuli including ligand-receptor interactions, growth factor deprivation, and inhibitors of cellular functions. All known caspases require cleavage adjacent to aspartates to liberate one large and one small subunit, which associate into an a2b2 tetramer to form the active enzyme. Caspase-14 has a conserved active site pentapeptide, QACRG. However, no proteolytic or biological activities have been identified so far. Its high expression in embryonic tissues and limited expression in adult tissues suggests that it may have some role during ontogenesis.

Eicosapentaenoyl serotonin is a hybrid molecule patterned after arachidonoyl serotonin (Item No. 70665). Arachidonoyl serotonin is a dual antagonist of fatty acid amide hydrolase (FAAH) and the transient receptor potential vanilloid-type 1 (TRPV1) channel, reducing both acute and chronic peripheral pain. The effects of replacing the arachidonoyl portion with eicosapentaenoic acid have not been studied. However, replacement of arachidonate with saturated 11- or 12-carbon fatty acids produces compounds that potently inhibit capsaicin-induced TRPV1 channel activation (IC50 = 0.76 μM) without blocking FAAH-mediated hydrolysis of arachidonoyl ethanolamine (IC50 > 50 μM).
