pTagGFP2-laminB1 vector

pTagGFP2-laminB1 is a mammalian expression vector encoding TagGFP2-lamin B1 fusion protein. The vector can be used for fluorescent labeling of lamin B1 in living cells. TagGFP2 codon usage is optimized for high expression in mammalian cells, i.e. humanized [Haas et al. 1996]. Human lamin B1 is fused to the TagGFP2 C-terminus. To increase mRNA translation efficiency, Kozak consensus translation initiation site is generated upstream of TagGFP2-lamin B1 coding sequence [Kozak 1987]. pTagGFP2-laminB1 vector can be used as a source of TagGFP2-lamin B1 hybrid sequence. The vector backbone contains unique restriction sites that permit its excision and further insertion into expression vector of choice.

Antibleaching live cell visualization medium DMEMgfp


Photobleaching of fluorescent proteins in response to prolonged exposure to exciting radiation significantly affects their utility as in vivo labels. The only ways of minimizing photobleaching known to date were limited to reducing of the excitation light intensity or shortening the duration of illumination. New Evrogen DMEMgfp live cell visualization medium excludes the components that lead to oxidative reddening of green fluorescent proteins, the photochemical process that was shown to be one of the main sources of GFP photobleaching [Bogdanov et al., 2009]. Replacing the culture medium with DMEMgfp for the period of visualization results in up to a 9-fold increase of photostability of EGFP, a 3.3-fold increase of photostability of TagGFP2 and more than a 4-fold increase of photostability of activated forms of photoactivatable PA-GFP and PS-CFP2 (the effect varies for particular fusions and localizations).
