Cell Migration Assays, 2D Gap Closure

Cell migration is a highly integrated, multi-step process that plays an important role in the progression of various diseases including cancer, atherosclerosis and arthritis. There are various types of cell migration which are defined here.

Cell invasion is related to, and encompasses, cell migration, except that cells do more than migrate. Invasive cells move through the extracellular matrix into neighboring tissues in a process that involves ECM degradation and proteolysis.

We offer cell migration assays in two formats:

  • Boyden Chamber Assays consist of a cell culture insert nested in the well of cell culture plate. Cells are seeded into the insert and move through the pores of the membrane at the bottom of the insert.
  • Gap Closure Assays create a defined area across which migratory cells can move. Migration can be monitored in real time by microscopy. These assays include our new proprietary Radius™ technology which uses a biocompatible hydrogel to create a circular area across which cells can migrate, and our Wound Healing Assay which is a more consistent alternative to the traditional scratch assay.

Tumor Sensitivity Assay

Traditionally, the soft agar colony formation assay has been used to monitor anchorage-independent growth. Cells proliferate for 3-4 weeks in a semisolid culture medium, followed by tedious manual counting of colonies.

Our CytoSelect™ 96-Well In Vitro Tumor Sensitivity Assay provides a stringent, anchorage-independent model for chemosenstivity testing and screening of potential anticancer drugs. After just 6-8 days, cell colonies are quantified using a standard colorimetric microplate reader.

Cell Invasion Assays

The ability of malignant tumor cells to invade normal surrounding tissue contributes in large part to the morbidity and mortality of cancers. Cell invasion requires several distinct cellular functions including adhesion, motility, detachment, and extracellular matrix proteolysis.

Our CytoSelect™ Cell Invasion Assays utilize precoated inserts to assay the invasive properties of tumor cells. Invasive cells can be quantified in 96-well plates on a fluorescence plate reader, or in 24-well plates on either a standard microplate reader or a fluorescence plate reader. Inserts are precoated on the top of the membrane with your choice of ECM matrix gel (basement membrane), Collagen I, or Laminin I.

Chemotaxis Assays

Chemotaxis describes the movement of cells toward or away from a chemical stimulus in their enviroment. Cell chemotaxis plays a pivotal role in the progression of cancer and other diseases.

CytoSelect™ Cell Migration Assays are ideal for determining the chemotactic properties of cells. The kits utilize polycarbonate membrane inserts in either 24-well or 96-well plates. Membranes are available in 4 pore sizes to accommodate a variety of cell types and sizes:

  • 8 µm pore size is suitable for most cell types including epithelial cells, fibroblasts, and cancer cell lines
  • 5 µm pore size is ideal for monocytes / macrophages
  • 3 µm pore size is best for the smallest cells including neutrophils and other leukocytes
  • 12 µm pore size is suitable for astrocytes and other large or slow-moving cells
