TLR ligand recognition and binding leads to signaling responses which most often include NF-κB and/or interferon regulatory factors (IRF). Both pathways are major circuits driving activation of hundreds of genes, many of which are inflammatory mediators and immune response factors. There is a clear connection between TLRs and NF-κB/IRF signaling pathways in programming and driving specific normal adaptive immune responses as well as in the development and maintenance of inflammatory, autoinflammatory diseases and cancer.
