- Scientific DescriptionThe TATA-binding protein (TBP) plays acentral role in the assembly of mosteukaryotic transcription initiation complexes.TBP assembles with other proteins to formunique multimeric complexes for each of thethree different nuclear RNA polymerases.TBP has been cloned from a variety ofspecies and consists of two distinct domains.The C-terminal domain (~180 amino acids)is highly conserved among species,whereas the N-terminal domain variesconsiderably in length and sequence amongdifferent species. The conserved C-terminaldomain contains the DNA-binding region aswell as regions that interact with positive andnegative regulatory proteins. In human TBP,the non-conserved N-terminal domainincludes a stretch of glutamine residuesranging from 26 to 42. Several researchgroups have found an association betweenexpansion of this polyglutamine tract withrare forms of spinocerebellar ataxia andother neurodegenerative diseases including Huntingtons disease.
