    Interleukin 18, also known as interferon-gamma inducing factor (IGIF) and IL1 gamma, shares some biologic activities with IL12 and structural similarities with the IL1 family of proteins. It plays an important role in the T cell helper type 1 (Th1) response through its ability to induce IFN-gamma production in T cells and NK cells. IL18 is related to the IL1 family in terms of structure, receptor family and function. Structurally, IL18 and IL1-beta share primary amino acids sequences (signature sequence motif) and are similarly folded as all-beta pleated sheet molecules. Similar to IL1 beta, production of IL18 requires caspase 1 (interleukin 1-beta converting enzyme, ICE) to cleave the pro-domain from the precursor protein to produce active mature IL18. Gene expression and synthesis of TNF, IL1, Fas ligand and several chemokines are induced by IL18. As with IL1, IL18 participates in both innate and acquired immunity. IL18 actions are mediated by an IL18 receptor (IL18R1) complex comprised of a binding chain (IL18 R-alpha, also called IL1 receptor-related protein, IL1 Rrp) and a signal-transducing chain. Both chains are members of the IL1 receptor family. The IL18R1 complex recruits the IL1 R-activating kinase (IRAK) and TNFR associated factor 6 (TRAF6) which phosphorylates NF-kappa-B-inducing kinase (NIK) with subsequent activation of NF-kappa-B.
