CPI-360 is a highly selective EZH2 inhibitor with IC50 values of 0.5 nM and 2.5 nM for wt EZH2 and Y641N EZH2, repectively. CPI-360 increases EZH2 protein stability at 52°C in a time-dependent manner. CPI-360 reduces H3K27 methylation levels in a dose-dependent manner without affecting EZH2, SUZ12, and EED protein levels.
