Cell Migration, Invasion and Wound Healing

Cell migration is a highly integrated, multi-step process that plays an important role in the progression of various diseases including cancer, atherosclerosis and arthritis. There are various types and definitions of cell migration.

Cell invasion is related to, and encompasses, cell migration, except that cells do more than migrate. Invasive cells move through the extracellular matrix into neighboring tissues in a process that involves ECM degradation and proteolysis.

Cell Biolabs offers cell migration assays in two formats:
  • Boyden Chamber Assays consist of a cell culture insert nested in the well of cell culture plate. Cells are seeded into the insert and migrate through the pores of the membrane at the bottom of the insert.
  • Gap Closure Assays create a defined area across which cells migrate. Cell migration can be monitored in real time by microscopy. These assays include Cell Biolabs new proprietary Radius™ technology which uses a biocompatible hydrogel to create a circular area across which cells can migrate, and their Wound Healing Assay which is a more consistent alternative to the traditional scratch assay.

Gap Closure Migration Assays

Cell Biolabs Gap Closure Assays provide a simple, convenient format to monitor cell migration with precision and accuracy. Each of these assays provides a consistently defined area across which migratory cells can move. Migration can be monitored in real time by microscopy.

Radius™ Cell Migration Assays

When cells are seeded in the well of the Radius plate, they will adhere everywhere except in the center of the well where we have placed a biocompatible hydrogel spot. Once cells form a monolayer, the assay is initiated by gently dissolving the gel with a removal solution, leaving a gap across which cell migration can occur.

Boyden Chamber Assays

The Boyden chamber is a useful tool to study cell migration and cell invasion. It consists of a cylindrical cell culture insert nested inside the well of a cell culture plate. The insert contains a polycarbonate membrane at the bottom with a defined pore size.

Cells are seeded in the top of the insert in serum-free media, while serum or similar chemoattractants are placed in the well below. Migratory cells move through the pores toward the chemoattractant below and can be stained or quantified in a plate reader. Invasive cells may be similarly measured by the placement of a coating of extracellular matrix proteins on top of the membrane.

Cell Biolabs CytoSelect™ Boyden Chamber Assays may be used to measure cell invasion as well as various types of cell migration:
  • Chemotaxis describes cell migration based on chemicals in a cell's surrounding environment. Chemotaxis can indicate cell migration either toward or away from a particular chemical signal.
  • Haptotaxis is defined as cell migration along a gradient of extracellular matrix-bound chemoattractants. In these assays the Boyden chamber insert is coated on the underside with Collagen I or Fibronectin protein.
  • Transmigration describes the migration of cells (usually leukocytes or tumor cells) through the vascular endothelium toward a chemoattractant.

Cell Invasion Assays

Cell Biolabs Cell Invasion Assays use a Boyden Chamber in which the top side of the 8 µm-pore membrane is coated with one of 3 gel layers:
  • Basement Membrane, a protein matrix isolated from Engelbreth-Holm-Swarm tumor cells
  • Collagen I
  • Laminin

Each Cell Invasion Assay kit is available in both 24-well and 96-well plate formats and contains a complete detection system for quantitation of invasive cells. No manual cell counting is required.

Chemotaxis Assays

Cell Biolabs Chemotaxis Assays use a Boyden Chamber containing a membrane with a defined pore size. Cells are seeded above the membrane and are allowed to migrate toward a chemoattractant in the lower chamber.

For optimal results the membrane pore size should be somewhat smaller than the size of the cell to be assayed. Use the following table to select the right pore size for your migration experiments.

Haptotaxis Assays

Haptotaxis is a type of cell migration defined by the migration of cells along a gradient of ECM-bound chemoattractants. Cell Biolabs Haptotaxis Assays are supplied with a Boyden Chamber containing 8 µm inserts that are precoated on the underside with either Collagen I or Fibronectin. Migratory cells are quantified using either a colorimetric or fluorescence plate reader.

Transmigration Assays

Transmigration involves the migration of cells across the endothelium. Cell Biolabs Transmigration Assays measure the interactions with, and transmigration of, the endothelium by leukocytes or tumor cells. The assays use a Boyden Chamber in which endothelial cells may be seeded to form a monolayer prior to adding the migratory cells of interest.
