The antigens are carefully designed, using a proprietary software to select the most suitable antigen for a given target protein. To secure that the very highest levels of specificity, reproducibility and versatility are achieved, a unique antibody purification process using the recombinant antigen as affinity ligand is performed.

Already today their catalogue includes over 18,000 Triple A Polyclonals that target more than 75% of the human protein coding genes. They are regularly expanding their catalog with the addition of up to 2,000 new Triple A Polyclonals per year, to reach their goal of providing you with high performance antibodies to all protein coding genes.

Specificity

Triple A Polyclonals are carefully designed using aproprietary antigen design software which selects a Protein Epitope Signature Tag (PrEST) sequence of between 50 to 150 amino acids having the lowest possible identity to other human proteins.

The PrEST concept secures the highest level of specificity when used for polyclonal antibody production. While full-length recombinant proteins often increase the risk for off-target binding, the PrEST antigen effectively reduces unwanted specificity by avoiding regions with high regional or local sequence identity to other proteins.

Reproducibility

Developed by top Swedish scientists for the creation of the Human Protein Atlas, their carefully designed Triple A Polyclonals are produced using the same standardized production process to ensure consistency. Their unique antibody purification process, using the recombinant PrEST-antigen as the affinity ligand, then achieves the very highest level of reproducibility new production lots are tested in parallel with existing lots to ensure functionality and assay reproducibility in a real lab situation.

Each Triple A Polyclonal antibody has been used for IHC-staining of samples from 708 individuals originating from normal and cancer afflicted human tissues. Trained pathologists have evaluated the reliability of all IHC-staining patterns. In addition ICC-IF-based confocal microscopy has been used to study subcellular distribution patterns in selected cell-lines. Further, Triple A Polyclonals are characterized by Western Blot (WB), primarily in endogenous protein lysates from human tissues.

Versatility

Triple A Polyclonals combine optimal specificity with superior versatility across applications including IHC, IF and WB. While anti-peptide antibodies often generate too narrowly specific antibodies, the use of Atlas PrEST-antigens results in advanced polyclonals that combine optimal specificity and minimal off-target binding with superior epitope recognition across applications, protein states and species.
