For applications where bulk cytokine or antibody levels are insufficient, ELISpot provides the necessary single‑cell functional resolution. Whereas ELISA reports total analyte concentration, ELISpot quantifies the number of cells actively secreting a defined protein, making it particularly powerful for dissecting antigen‑specific responses, rare effector populations and functional T and B cell immunity.
How ELISpot Works: Technology & Sensitivity
The ELISpot assay is an extremely sensitive immunoassay in 96‑well format that detects secreted proteins captured on a membrane and visualized as discrete spots, each corresponding to one secreting cell. With the capacity to detect approximately one antigen‑specific cell in a million, ELISpot is ideally suited for monitoring low‑frequency T and B cell responses in infection, cancer, autoimmunity and allergy, as well as vaccine and cell therapy programs.
