Advancing Extracellular Vesicle Research: OriGene’s New EV Isolation Reagent and Exosome ELISA Kit

Extracellular vesicles (EVs), including exosomes ranging from 30-150 nm in size, are secreted by cells into biofluids such as blood, cerebrospinal fluid, and urine. These vesicles carry proteins, RNA, and DNA and are rapidly emerging as powerful biomarkers and therapeutic delivery vehicles. As interest in EV biology accelerates across oncology, neuroscience, and immunology, the demand for reliable, high-performance isolation and detection tools has never been greater.

Key advantages of the OriGene’s EV Isolation Reagent include:

In head-to-head comparisons, BCA protein quantification and Western blot analysis confirmed that OriGene’s reagent outperforms competing precipitation products (including Vendor Q), yielding stronger CD81 signals and greater total protein content from stem cell conditioned medium.

Importantly, Nanoparticle tracking analysis (NTA) demonstrated significantly higher EV recovery (>10-fold) using OriGene’s EV isolation reagent compared to ultracentrifugation.

For EV marker validation, Simple Western analysis of HEK293T-derived EVs demonstrated clear detection of CD63, TSG101, CD81, and MFGE8, all well-established exosomal markers.

The EV Isolation Reagent is available in two formats: 15 mL (SKU: AR100029s) and 50 mL (SKU: AR100029).

Sensitive Exosome Quantification with the CD9 ELISA Kit

Complementing the isolation reagent, OriGene’s new Human Exosome ELISA Kit (CD9 Detection, SKU: EA200081) enables direct quantification of CD9-positive exosomes from native biofluids, without requiring a prior isolation step. This minimizes EV loss and simplifies the quantification workflow.

The kit delivers exceptional analytical performance:

  • Sensitivity (LLOD): 1.91 × 10⁶ particles/mL (approximately 13.2 ng/mL total exosome protein equivalence)
  • Assay Range: 4.53 × 10⁶ to 2.9 × 10⁸ particles/mL
  • Average Spike Recovery: Cell culture (95%), plasma (109%), serum (96%)
  • Precision: Intra-assay CV <6%; Inter-assay CV <4%

The standard curve remains highly linear down to approximately 2.9 × 10⁸ particles/mL, and typical sample types: serum, EDTA plasma, and cell culture media, all fall within the assay’s dynamic range, making it broadly applicable across research settings.
