Plant RNA extraction can be difficult, especially when working with tissues rich in polysaccharides, polyphenols, and other compounds that may interfere with RNA purification. Leaves, roots, stems, seeds, fruits, soybeans, corn, and other complex plant materials often require a workflow that is both robust and practical for routine laboratory use.
The ARPENspin Plant Total RNA Extraction Kit from ARPENGEN is designed to simplify total RNA isolation from plant tissues, including challenging samples containing polysaccharides and polyphenols. The kit provides a spin-column workflow that supports high-quality RNA purification through sample lysis, column binding, washing, DNase I treatment, and elution.

Different plant samples do not always behave the same during RNA extraction. To support this, the kit includes two lysis buffer options, Lysis AG and Lysis BG, as well as PLANTaid for more demanding samples. According to the protocol, Lysis AG can be used for most plant samples, while Lysis BG or Lysis BG with PLANTaid may be selected when extraction results need improvement.
This flexible lysis approach makes the kit especially relevant for laboratories working with diverse plant materials or sample types that are known to be difficult to process.
The workflow is designed to be straightforward and efficient. After plant tissue is ground with liquid nitrogen and mixed with the selected lysis buffer, the lysate is clarified by centrifugation and transferred to a spin column. RNA is then purified through washing steps, on-column DNase I treatment, and final elution.
The flyer describes the process as an approximately 40-minute workflow and highlights recovery rates reaching up to 90%. The protocol also avoids phenol, chloroform, and other organic solvents, helping create a safer and more user-friendly RNA extraction process.
The purified RNA is suitable for a wide range of downstream applications, including Northern blotting, hybridization, poly(A)+ selection, in vitro translation, RNase protection assays, RT-PCR, real-time RT-PCR, and cDNA library construction. The data sheet also lists sequencing, mutant analysis, SNP analysis, and other applications.
