LymphoTrack Assays & Workflow

Invivoscribe offers a suite of research use only (RUO) kits targeting seven common gene rearrangements—including IGH (Leader and FR1/2/3), IGK, TRB and TRG—to support clonality identification, somatic hypermutation determination and measurable residual disease (MRD) assessment.  All assays feature indexed master mixes and a streamlined one-step multiplex PCR that allows up to 24 samples and any combination of targets per run.  Resulting FASTQ files are analyzed by the RUO LymphoTrack Software – MiSeq* and/or  LymphoTrack MRD Software* which generates PDF reports and tabulated output files for result interpretation and intuitive data visualization.

Comparative Performance: MiSeq vs. MiSeq i100

Invivoscribe constructed a seven-target library from contrived clonal and polyclonal DNA dilutions to compare the original MiSeq (with v3 Standard-SBS chemistry) against Illumina’s MiSeq i100 (using XLEAP-SBS chemistry). Key findings include:

• Run Time: MiSeq i100 completed 2×300 bp paired-end sequencing in ~14 hours—nearly 4x faster than the ~53 hours needed on MiSeq.
• Output & Quality: Both systems generated similar data volumes (32–34 million pass-filter reads; ~20 Gb output). However, the MiSeq i100 achieved a higher Q30 score (96.9% vs. 77.0%), reflecting enhanced sequencing accuracy.
• Data Concordance: All targets exceeded the 20,000-read validity threshold on both platforms, and V- and J-gene assignments were fully concordant.  Top-percent reads for each clonal sequence were virtually identical (R² > 0.99 across all seven targets).
