Cell Guidance Systems launches Instant Exosomes™ and LipoQ™ assay

Cell Guidance Systems launches two new products: Instant Exosomes™, a range of well characterized freeze-dried exosomes and LipoQ™, a lipid quantification assay


Instant Exosomes™

Exosomes are a subtype of Extracellular vesicle (EV), lipid membrane-enclosed compartments ranging in size from 50- to 1000 nm in diameter. EVs are released from every cell type and tissue, and their contents are incredibly diverse. They contain cellular components such as proteins, bioactive lipids, and nucleic acids originating from the releasing cell. Exosomes transport these molecules between cells, and are therefore important in cell signalling processes.

Cell Guidance Systems is now offering a range of high-quality and well-characterized freeze-dried exosome samples from human cancer cell lines for research applications. The freeze-dried exosomes are isolated using Exo-spin™ kits following a combination of precipitation and size exclusion chromatography (SEC).

Each batch of freeze-dried exosomes is quality tested and guaranteed to be of the highest standard. The exosome samples are characterized for:

  • Expression of commonly expressed exosomal markers (CD9, CD63 and CD81) using the ExoLISA™ exosome detection assay. Each sample expresses the commonly expressed exosomal markers CD9, CD63 and CD81. This also ensures that other surface proteins that are present should be detectable.
  • Overall protein content using a BCA assay.
  • Particle concentration and size distribution by nanoparticle tracking analysis (NTA) using a Zetaview® instrument from Particle Metrix GmbH.

These samples are freeze-dried to maximise their stability for storage and shipping, enabling these products to be stored at 4°C until reconstituted. Exosome samples are simply reconstituted by adding water. Exosome samples are available from widely used human cancer cell lines. There is a wide and diverse range of applications for this range of freeze-dried exosome samples, from assay calibration to protein marker analysis.

LipoQ™

Lipids represent a large and diverse group of molecules, which can be divided into eight basic groups: fatty acyls, glycerolipids, glycerophospholipids, sphingolipids, sterol lipids, prenollipids, saccharolipids, and polyketides. Not only are lipids key components of cellular compartments, but they also store energy and have important signalling roles. In order to study lipids, they must often be extracted first from tissues or cellular cultures and then quantified. There are various methods available for lipid quantification, ranging from state-of-the-art quantification using advanced technology to simpler benchtop solutions.

LipoQ™ is based on the sulfo-phospo-vanillin method, which measures the lipid content of unsaturated fatty acids within a sample. Unsaturated fatty acids within a sample will react with concentrated sulphuric acid, which in combination with phospho-vanillin will form a pink-colored solution. The intensity of the pink colour formed is determined by the total lipid concentration within the sample, enabling a reliable colorimetric assay that can be read using a plate reader. Cell Guidance Systems has developed the LipoQ™ lipid quantification kit based on this principle. This simple, benchtop top assay is amenable to a multiwell plate and results in a simple colorimetric readout, that does not require the use of equipment not usually available in research laboratories.
