1. Prepare Protein Solution: Dissolve 10 mg of your protein in 0.1 M sodium bicarbonate buffer.
2. Dye Preparation: Dissolve NHS ester in DMF or DMSO at 10 mg/mL immediately before use.
3. Conjugation: Add 50–100 uL of AbBy Fluor NHS Ester solution to the protein solution while stirring; incubate 1 hour at room temperature.
4. Purification: Separate labeled conjugate from free dye using a gel-filtration column (Sephadex® G-25, BioGel® P-30, or equivalent).
5. Characterization: Measure absorbance at 280 nm and dye λmax to calculate degree of labeling (DOL).
