One-Hour Fluorescent Labeling—No Kits Needed

Turn any protein, antibody, or biomolecule into a vibrant, photostable fluorescent probe in just one hour—no specialty kits, enzymes, or reagents required. Eight dyes span 350–750 nm for seamless panel design.

AbBy Fluor™ NHS Esters: Rapid, Versatile, and Cost-Effective Fluorescent Labeling

In the fast-paced world of life-science research, every minute of bench time counts. AbBy Fluor™ NHS Esters allow researchers to transform any primary-amine–containing biomolecule into a bright, long-lasting fluorescent probe in a simple, single-step reaction that takes just one hour at room temperature. By pre-activating the N-hydroxysuccinimide (NHS) ester group on our dyes, we eliminate the need for enzymes, click chemistry reagents, or proprietary consumable kits—saving both time and cost while maximizing experimental flexibility.

Key Advantages: AbBy Fluor™ NHS Esters

1. One-Hour, One-Step Protocol

Simply dissolve your protein or antibody in sodium bicarbonate buffer, add the AbBy Fluor™ NHS ester dissolved in DMF or DMSO, and incubate for one hour. No further kits or enzyme mixes are required, streamlining your workflow.

2. Eight Distinct Dyes (350–750 nm)

From ultraviolet (350 nm excitation) through near-infrared (750 nm excitation), our eight-member dye family lets you design complex multi-color panels on any common confocal, flow cytometry, or widefield fluorescence setup—without worrying about compatibility or spectral overlap.

3. Cost-Effective Labeling

AbBy Fluor™ NHS Esters offer significant savings compared to purchasing large volumes of pre-labeled antibodies and reduce the need to stock multiple conjugates.

4. Broad Application Range

  • Immunofluorescence (IF) & Immunohistochemistry (IHC): Generate high-contrast antibody probes that retain antigen affinity and signal stability.
  • Flow Cytometry (FCM): Create custom conjugates tailored to your panel design, even for rare fluorophore combinations.
  • Ligand & Peptide Tracking: Label small ligands or peptides for real-time imaging of receptor binding and trafficking.
  • FRET & Biosensor Pairs: Mix and match donor–acceptor combinations for quantitative energy transfer assays and live-cell sensor construction.
  • Surface & Bead Functionalization: Coat microbeads or sensor surfaces with fluorescent tags for high-throughput screening.

Protocol Overview

1. Prepare Protein Solution: Dissolve 10 mg of your protein in 0.1 M sodium bicarbonate buffer.

2. Dye Preparation: Dissolve NHS ester in DMF or DMSO at 10 mg/mL immediately before use.

3. Conjugation: Add 50–100 uL of AbBy Fluor NHS Ester solution to the protein solution while stirring; incubate 1 hour at room temperature.

4. Purification: Separate labeled conjugate from free dye using a gel-filtration column (Sephadex® G-25, BioGel® P-30, or equivalent).

5. Characterization: Measure absorbance at 280 nm and dye λmax to calculate degree of labeling (DOL).
