The ongoing pandemic of the newly identified COVID-19 has been declared a public health emergency of international concern by the WHO. Several of our suppliers offer products to assist the research related to this virus.

The causative SARS-CoV-2 virus attaches to cells by binding its outer “Spike” proteins to a receptor protein called ACE2 (angiotensin-converting enzyme 2) that is highly expressed on respiratory cells. ACE2 also acts as the receptor for the human respiratory coronavirus NL63 and the SARS-coronavirus (SARS-CoV). Studies have shown that the binding affinity of the virus Spike protein for ACE2 correlates with higher transmissibility of the coronavirus and contagiousness of the disease. Angiotensin receptor blockers have been proposed as tentative SARS-CoV-2 therapeutics. Interestingly, ACE2 activity may also have a protective effect against virus-induced lung injury by increasing the production of the vasodilator angiotensin 1–7. [ref. 1-5]

COVID-19 relevant tools by Sino Biological

Sino Biological successfully expressed COVID-19 spike proteins to develop therapies or further research the virus. This new virus belongs to the Betacoronavirus Genus, which also includes SARS CoV (2003) and MERS CoV (2012). The sequence reveals that COVID-19 is more similar to SARS-CoV (70% similarity) than to MERS-CoV (40%). Same as all other coronaviruses, the genome of COVID-19 encodes the spike protein, the envelope protein, the membrane protein, and the nucleocapsid protein.

The spike protein (S-protein)

This protein mediates receptor binding and membrane fusion. Spike protein contains two subunits, S1 and S2. S1 contains a receptor binding domain (RBD), which is responsible for recognizing and binding with the cell surface receptor. S2 subunit is the "stem" of the structure, which contains other basic elements needed for the membrane fusion. The spike protein is the common target for neutralizing antibodies and vaccines. It’s been reported that COVID-19 can infect the human respiratory epithelial cells through interaction with the human ACE2 receptor. Indeed, the recombinant Spike protein can bind with recombinant ACE2 protein [Xu et al., 2020].
The nucleocapsid Protein (N-protein) is the most abundant protein in coronavirus. The N-protein is a highly immunogenic phosphoprotein, and it is normally very conserved. The N protein of coronavirus is often used as a marker in diagnostic assays.

The GTX632604 antibody was raised against SARS-CoV spike protein, and based on the feedback it detects SARS-CoV-2 in infected cells as well by Western blot, therefore the product name is changed to SARS-CoV/ SARS-CoV-2 (COVID-19) spike antibody [1A9]. The data is available on the datasheet. In addition, GeneTex also has an antibody against ACE2 (GTX101395), the host receptor for SARS-CoV and SARS-CoV-2 to enter host cells.

SARS-CoV-2 (COVID-19) spike antibody (GTX135360) and SARS-CoV-2 (COVID-19) nucleocapsid antibody (GTX135361) are both raised against a peptide antigen of which the regions are not identical in SARS-CoV-2 and SARS-CoV.

COVID-19 poses a serious threat to people's health, and it is urgent to develop drugs to treat COVID-19 quickly. The screening of anti-COVID-19 drugs by using the clinical and approved compounds can greatly shorten the research and development cycle. In addition, the virtual screening technology can effectively narrow the scope of screening and improve the screening efficiency in the pre-screening of new drugs.

The causative SARS-CoV-2 virus attaches to cells by binding its outer “Spike” proteins to a receptor protein called ACE2 (angiotensin-converting enzyme 2) that is highly expressed on respiratory cells. ACE2 also acts as the receptor for the human respiratory coronavirus NL63 and the SARS-coronavirus (SARS-CoV). Studies have shown that the binding affinity of the virus Spike protein for ACE2 correlates with higher transmissibility of the coronavirus and contagiousness of the disease. Angiotensin receptor blockers have been proposed as tentative SARS-CoV-2 therapeutics. Interestingly, ACE2 activity may also have a protective effect against virus-induced lung injury by increasing the production of the vasodilator angiotensin 1–7. [ref. 1-5]

Biotium: A closed-tube, diagnostic test for COVID-19 at home, clinic, and points of entry

Current methods for qualitative detection of COVID-19 primarily depend on reverse transcription-PCR (RT-PCR). However, RT-PCR tests can produce false negative results due to insufficient detection sensitivity for patients with a low viral load. Moreover, RT-PCR requires well-equipped laboratories and skilled personnel. Due to the rapid spread of the virus and limited capacity of hospitals, a more accurate, rapid, and easy-to-use molecular diagnostic for COVID-19 is urgently needed.

In a recent preprint manuscript posted on ChemRxiv™, El-Tholoth and colleagues at the University of Pennsylvania have developed simple, point-of-care assays for qualitative COVID-19 detection using one-stage LAMP (loop-mediated isothermal amplification) or two-stage Penn-RAMP (recombinase polymerase amplification plus LAMP). Specificity was accomplished by amplifying a conserved sequence of ORF1ab from the COVID-19 genome with high homology to the virus and high divergence from all other coronaviruses. The one-stage LAMP assay contains an isothermal master mix with EvaGreen® dye for reliable and robust amplification. Results show the one-stage LAMP assay performs comparably to RT-PCR methods for COVID-19 detection in tests with purified template and mock patient samples. Results for the two-stage Penn-RAMP assay showed detection sensitivity of 100 times higher than the one-stage LAMP or RT-PCR methods in similar mock samples. Moreover, the Penn-RAMP assay can be performed entirely in a closed tube and is suitable for colorimetric detection by LCV (leuco crystal violet). Given the simplicity, high-sensitivity, and minimal instrumentation required by Penn-RAMP, the authors propose further validation for its use as a point-of-care COVID-19 diagnostic.

Viral Transport Media by Life Science Group

Life Science Group has announced that they are now manufacturing Viral Transport Medium to both CDC and WHO protocols.

The constituents of suitable viral transport media are designed to provide an isotonic solution containing protective protein, antibiotics to control microbial contamination, and one or more buffers to control the pH.

Liquid transport media are used primarily for transporting swabs or materials released into the medium from a collection swab. Liquid media may be added to the specimens when inactivation of the viral agent is likely and when the resultant dilution is acceptable.
